AKT proteins are activated by phosphorylation via phosphoinositide 3-kinase (PI3K) and play key roles in various signaling pathways, including those involving receptor tyrosine kinases and G-protein coupled receptors. AKT activation requires phosphorylation of threonine 308 and serine 473. The PI3K/AKT pathway is crucial for tumor cell survival by inhibiting apoptosis and activating survival factors. AKT also interacts with the mTOR pathway, affecting translation initiation and is often dysregulated in cancers. Mutations in this gene are linked to various cancers, Proteus syndrome, Cowden syndrome, and other growth disorders. Multiple transcript variants exist due to alternative splicing.

This protein kinase lies upstream of MAP kinases and stimulates the enzymatic activity of MAP kinases upon wide variety of extra- and intracellular signals. As an essential component of MAP kinase signal transduction pathway, this kinase is involved in many cellular processes such as proliferation, differentiation, transcription regulation and development.
